Saffron

The most-studied culinary spice for mood: multiple head-to-head RCTs vs fluoxetine in mild-to-moderate depression.

Why

Saffron (Crocus sativus stigmas) has become one of the better-studied herbal antidepressants of the last decade. Meta-analyses of more than 20 RCTs report effects comparable to fluoxetine and imipramine in mild-to-moderate depression, including direct head-to-head trials. Trials are mostly from Iranian centres and short in duration, but the consistency of direction is unusual for a herbal product.

How it works

Crocins and safranal inhibit reuptake of serotonin, dopamine and noradrenaline, modulate the HPA axis, and show NMDA-receptor antagonist activity in preclinical work. Antioxidant effects on hippocampal neurons are documented mechanistically.

How to take

Dosage

30 mg standardised extract daily, typically divided 15 mg twice daily, the dose used in the majority of trials.

Look for standardised extracts such as affron® or Satiereal® with stated crocin/safranal content. Whole-stigma capsules vary widely.

Ideal for

Adults with mild-to-moderate low mood looking for an option with fewer interactions than St John's wort.

Safety

Doses above 1.5 g/day are toxic; stay within standardised supplement ranges. Avoid in pregnancy (uterotonic at high doses). Theoretical additive serotonergic effects with SSRIs and SNRIs, so use caution if combining with prescribed antidepressants.

Tóth 2019 Planta Med meta-analysis (23 RCTs, n=1,499): standardised saffron extract was significantly more effective than placebo and similar in efficacy to first-line antidepressants (fluoxetine, citalopram, imipramine) for mild-to-moderate depression over 6–12 weeks. Effect size moderate; trial quality variable but consistent in direction.
