Lion's mane (Hericium erinaceus)

Edible medicinal mushroom with preliminary RCT signal for mild cognitive complaints and an NGF-promoting mechanism.

Why

Lion's mane (Hericium erinaceus) is an edible mushroom that has attracted attention for its hericenone and erinacine content, compounds that stimulate nerve growth factor (NGF) in vitro and in animal models. A small Japanese RCT in older adults with mild cognitive impairment reported improved cognitive scores at 16 weeks. Other small trials suggest effects on mood and gastric mucosal protection. Evidence is genuinely preliminary: much smaller body than ginkgo or bacopa.

How it works

Hericenones (fruiting body) and erinacines (mycelium) cross the blood-brain barrier in animal models and stimulate NGF synthesis, supporting neuronal differentiation and survival. Independent effects on gastric mucosa and gut microbiota are documented in preclinical work.

How to take

Dosage

Most trials use 1,000–3,000 mg/day of fruiting-body extract divided across the day. Standardised erinacine A-enriched mycelium extracts are dosed differently; follow product label.

Timing

With meals; divided 2–3 times daily

On the label

Specify 'fruiting body' (where hericenones concentrate) or 'erinacine A-enriched mycelium'. Avoid 'mycelium on grain' products which are mostly grain by weight.

Adults exploring early-evidence cognitive-support options; those with mild subjective memory complaints under clinical guidance.

Safety

Allergic reactions and skin rashes have been reported, particularly in those with mushroom or mould allergies. Theoretical antiplatelet effect at high doses; caution with anticoagulants. Pregnancy and breastfeeding data limited. Variable product quality is the practical limit.

Mori 2009 Phytother Res small RCT (n=30 Japanese adults with MCI): Hericium 1,000 mg three times daily improved cognitive function scores at 8, 12 and 16 weeks vs placebo, with effect lost 4 weeks after cessation. Modern evidence base is still small. Cognitive signal is real but preliminary; mechanism via NGF is well documented preclinically.
